Hyperosmotic shock induces cytochrome c release and capase-3 activation in Xenopus oocytes, but the regulators and signaling pathways involved are not well characterized. Here we show that hyperosmotic shock induces rapid calpain activation and high levels of Smac/DIABLO release from the mitochondria before significant amounts of cytochrome c are released to promote caspase-3 activation. Some properties of signaling systems, like ultrasensitivity, hysteresis (a form of biochemical memory), and all-or-none responses at a single cell level, are important to understand the regulation of irreversible processes. [...]

Ionizing radiation damages DNA and induces mutations as well as chromosomal reorganizations. Although radiotherapy increases survival among cancer patients, this treatment does not come without secondary effects, among which the most problematic is gonadal dysfunction, especially in women. [...]

Impaired development of embryos produced by somatic cell nuclear transfer (SCNT) is mostly associated with faulty reprogramming of the somatic nucleus to a totipotent state and can be improved by treatment with epigenetic modifiers. [...]

14 p, 213.3 KB Effect of Roscovitine on nuclear maturation, MPF and MAP kinase activity and embryo development of prepubertal goat oocytes / Jiménez y de Macedo, Ana Raquel (Universitat Autònoma de Barcelona. Departament de Ciència Animal i dels Aliments) ; Izquierdo Tugas, Ma. Dolors (Maria Dolors) (Universitat Autònoma de Barcelona. Departament de Ciència Animal i dels Aliments) ; Urdaneta Vargas, Aixa Efrailda (Universitat Autònoma de Barcelona. Departament de Ciència Animal i dels Aliments) ; Paramio Nieto, María Teresa (Universitat Autònoma de Barcelona. Departament de Ciència Animal i dels Aliments)
The low number of embryos obtained from IVM–IVF–IVC of prepubertal goat oocytes could be due to an incomplete cytoplasmic maturation. Roscovitine (ROS) inhibits MPF and MAP kinase activity and maintains the oocyte at Germinal Vesicle (GV) stage. [...]
