Large-scale production of adeno-associated viral vectors for human gene therapy : Alzheimer's disease. Part II: Upstream and Bioreaction

Abstract: Currently, one of the main bottlenecks in the development of many human gene therapies is the lack of large-scale production methods of the required vectors. Owing to this fact, this work focuses on the development of an adeno-associated viral vector production process, specifically serotype 9 (AAV9), using suspension human embryonic kidney cells (HEK293) for the treatment of Alzheimer's disease. The proposed production process, based on perfusion culture in a rocking motion bioreactor, proves to be a robust method, achieving high yields greater than 2·10^5 vg/cell in serum-free conditions and without the need to use helper virus due to the triple plasmid transfection method using polyethylenimine (PEI).
