Abstract: La presència de metalls de transició a la natura no ve donada per la mà de l'home. De fet, les propietats electròniques i estructurals d'aquestes peces centrals de la taula periòdica són clau per la regulació dels processos biològics tals com el transport d'oxigen en animals o la fotosíntesi a les plantes. Les àrees de la farmacologia, la química, la medicina i la biotecnologia han après les lliçons d'organometàl·lica impartides per la natura, fent evolucionar la seva recerca cap a processos més específics. En aquesta tesi, titulada "Predicció d'interaccions biometàl·liques: reptes i aplicacions" ("Prediction of biometallic interactions: challenges and applications"), s'han centrat tots els esforços per descriure els diferents efectes estructurals i electrònics del acoblament d'un compost organometàl·lic sobre un sistema biològic (ADN, proteïnes i pèptids) aplicant tècniques de modelització molecular. Existeix un gran ventall de tècniques de modelització molecular. Escollir una o altre no només depèn del sistema sobre el qual s'ha d'aplicar, sinó que també s'ha de tenir en compte la interacció química que succeeix. Bàsicament, les tècniques de modelització molecular es poden dividir en dos: aquelles basades en els principis de la mecànica quàntica i les que es descriuen segons els fonaments de la mecànica molecular. Els mètodes de mecànica quàntica son capaços de descriure de forma precisa els efectes electrònics dels metalls, però consumeixen més temps de computació que els de mecànica molecular, essent aquests la millor opció per estudiar sistemes grans. Aquesta tesi està composta per quatre capítols de resultats que mostren la feina feta durant els últims anys dedicats a la recerca. En tots quatre s'han fet servir diferents tècniques de modelització molecular per estudiar diferents processos químics i biològics que involucren la participació de metalls de transició. En aquests capítols s'ha investigat en 1) l'ús de tècniques d'acoblament proteïna-lligand fent servir lligands organometàl·lics, 2) la descripció de l'estat de repòs i del procés d'activació d'un metal·loenzim artificial fent servir el mètode híbrid de Mecànica Quàntica/Mecànica Molecular, 3) l'estudi dels efectes a curt i llarg abast de la coordinació de cisplatí en ADN i proteïnes combinant les tècniques de mecànica quàntica i anàlisi de modes normals, i 4) en les idees inicials sobre la formació enantioselectiva de metal·lopèptids artificials combinant coneixements teòrics i experimentals. Escollir la millor tècnica a cada punt va ser la clau per descriure el sistema químic i descriure el corresponent procés biològic de forma satisfactòria.
Abstract: The presence of transition metals in nature is not a man-made fact. In fact, the structural and electronic properties provided by that particular pieces of the periodic table are key in the regulation of biological processes such as the oxygen transport in animals or the photosynthesis in plants. Pharmacology, chemistry, medicine and biotechnological fields learned the lessons of organometallic chemistry given by Nature, evolving their research to more specific processes. In this thesis, titled "Prediction of biometallic interactions: challenges and applications", all the efforts have been focused in the description of the structural and electronic effects of the binding of an organometallic compound to a biological host (DNA, proteins and peptides), by the application of molecular modelling techniques. The molecular modeller's toolbox includes a huge variety of methods to choose. The election of one of other not only depends on the system where it has to be applied, but also the chemical interaction is an important variable to take into account. Molecular modelling techniques can be divided in two fields: those based in the principles of quantum mechanics, and the other which are described using the fundamentals of molecular mechanics. Quantum mechanics methods accurately describes the electronic effect of the metal, but are more time consuming that molecular mechanics ones, which are the best option to study large systems. This thesis is composed by four chapters presenting the results obtained during the last years of research. In all four, different chemical and biological processes involving the interaction of transition metals and biological hosts have been studied using different computational tools. In these chapters we investigated on 1) the use of protein-ligand docking with organometallic ligands, 2) the description of the resting state and activation process of an artificial metalloenzyme using the hybrid Quantum Mechanics/Molecular Mechanics approach, 3) the study of the short and long range effects of the coordination of cisplatin in DNA and proteins mixing Quantum Mechanics and Normal Mode Analysis, and 4) on the clues on the enantioselective formation of artificial metallopeptides combining theoretical and experimental knowledge. The election of the best technique at each stage of research was key to describe the chemical system and succeed on the description of the involved biological process.
