Estudi de l'especificitat metàl·lica i del papers dels lligands sulfur en metal·lotioneïnes (MTS) de diversos organismes

Abstract: Les Metal·lotioneïnes (MTs) són unes metal·loproteïnes de baix pes molecular (60-100 aa) amb un elevat contingut en residus de cisteïna, les quals els confereixen la capacitat d'enllaçar metalls pesants. Ara bé, existeixen altres lligands tant proteics (histidines) com no proteics (sulfurs àcid-làbils i clorurs) que també poden participar en la coordinació metàl·lica. Si bé se les ha relacionat amb diferents funcions dins de l'organisme (detoxificació enfront de metalls pesants, homeòstasi de Zn i Cu, funcions antioxidants i antiapoptòtiques, etc), encara es treballa intensament per determinar la seva funcionalitat específica. Aquest fet ha dificultat la seva classificació, de manera que fins a l'actualitat se n'han fet diferents propostes. La més recent es centra en la preferència metàl·lica de les MTs envers els ions essencial Zn(II) i Cu(I). Aquests ions, amb comportaments coordinants clarament diferenciats, probablement determinen estructures diferents per als complexos M-MT, les quals, tenint en compte la relació estructura/funció, segurament porten a funcions diferenciades per a les anomenades Zn- i Cu-tioneïnes. Amb l'objectiu d'aprofundir en el coneixement actual d'aquestes proteïnes, en aquesta Tesi Doctoral s'han estudiat un elevat número de MTs de diversos organismes, tant d'animals (nematodes i mol·luscos) com de planta (alzina surera) i de llevat. Els treballs realitzats amb les metal·lotioneïnes CeMT1 i CeMT2 del nematode C. elegans han permès conèixer amb més detall el paper que juguen els aminoàcids histidina en la coordinació metàl·lica en les MTs i com aquests aminoàcids determinarien el caràcter de Zn-tioneïna d'una MT. Els treballs realitzats amb la MT de planta (QsMT de l'alzina surera Q. suber) han permès aprofundir en el coneixement actual sobre el paper dels lligands sulfur àcid-làbils en les MTs recombinants, observant-se que aquests lligands augmenten la capacitat coordinant de QsMT enfront el Cd(II). Per altra banda, gràcies a la purificació d'una MT nativa de llevat (Cup1) s'ha demostrat que els agregats metall-MT d'aquesta proteïna també contenen anions sulfurs àcid-làbils, de manera que aquests lligands es troben presents en les metal·lotioneïnes natives i per tant no són un artefacte de la síntesi recombinant a partir de cèl·lules de bacteri, tal i com alguns especialistes en el camp havien proposat. Un dels punts forts d'aquesta Tesi Doctoral ha estat l'estudi de la reactivitat de les MTs enfront els radicals reductors, on s'ha demostrat per primera vegada que els radicals reductors poden causar la desulfurització dels aminoàcids cisteïna i metionina de les MTs causant danys a les membranes cel·lulars. Per últim, gràcies a l'estudi d'un considerable nombre de MTs de diversos organismes, s'ha pogut refinar la classificació anterior de les MTs per part d'aquest grup de recerca en Zn-tioneïnes i Cu-tioneïnes, demostrant que existeix una gradació entre aquests dos grups, formant un grup amb característiques intermèdies que podrien comportar-se com a Cu- o Zn-tioneïnes depenent de les necessitats específiques de la cèl·lula.
Abstract: Metallothioneins (MTs) are low molecular weight proteins (60-100 aa) with a high cysteine content (30% of the total amino acid content), which confers them the ability to bind heavy metal ions. As well as the Cys residues, other kind of proteic (histidines) and non-proteic (acid-labile sulfide and chloride) ligands exists in MTs that also could participate in metal coordination. Although they have been related to different functions in the organism (heavy metal detoxification processes, Zn and Cu homeostasis, antioxidant and antiapoptotic functions, etc), their physiological function is still a matter of debate and prompts most of the research within this field. This fact has difficulted their classification, and in consequence several models of classification have been proposed. The most recent one is based on the metallic preferences of the MTs towards the essential Zn(II) and Cu(I) ions. These ions clearly show different coordinative abilities and must, therefore, determine the formation of different M-MT structures. Thus, when considering the structural/function relationship, these differences in the structure of the M-MT complexes might determine different functions for the so-called Zn- and Cu-thioneins. Thus, and with the objective of increasing the actual knowledge about these proteins, the Zn(II), Cd(II) and Cu(I) coordinative abilities of a large number of MTs from different organisms, such as animals (nematode and mollusk), plants (cork oak) and yeast have been studied in this PhD. The work performed with CeMT1 and CeMT2 from the nematode C. elegans has shed light on the role played by the histidines in the metallic coordination in MTs and how these amino acids are responsible for determining the Zn-thionein nature of MTs. The work performed with the plant MT (QsMT, from the cork oak Q. suber) has allowed to increase the actual knowledge about the role of the acid-labile sulfide ligands in recombinant MTs, noticing that these ligands increase the coordinative abilities of QsMT when binding Cd(II). On the other hand, thanks to the purification of a native MT from yeast (Cup1), it has been demonstrated that the metal-MT complexes from this protein contains acid-labile sulfide anions, and therefore these ligands are also present in native metallothioneins, and therefore, they are not an artifact from the recombinant synthesis in bacteria cells, just as some specialists in the field proposed. One of the strong points of this Thesis has been the study of the MTs' reactivity against the reductive radicals, where it has been demonstrated by first time that this type of radical species could cause the desulfurization of the metionine and cysteine amino acids, causing damage to the cell membrane in turn. Finally, thanks to the study of a considerable number of MTs from different organisms, it has been possible to refine the MT classification previously presented by this group in Zn- or Cu-thioneins. Thus, it has been demonstrated that a gradation between these two extreme groups of MTs exists, which leads to a group of proteins with intermediate features that could behave as Cu-thioneins or Zn-thioneins depending on the specific requirements of cell.
