Targeting Porphyromonas gingivalis with ginger exosome-like nanoparticles : the next cure of Alzheimer?

Alzheimer's type dementia (AD) is a debilitating neurodegenerative disorder affecting 55 million people globally. It is characterised by the presence of extracellular senile plaques and intracellular neurofibrillary tangles, consisting of amyloid beta and hyperphosphorylated tau proteins respectively. Despite global research efforts, there is currently no cure available. Porphyromonas gingivalis is a risk factor of the development of this disease. Indeed, this bacteria infects more than half of the world's population. Consequently, controlling P. gingivalis infection is key to fight against AD. However, this bacterium is resistant to broad-spectrum antibiotics; thus, a new therapy is needed to inhibit the deleterious effects of this bacteria on the brain. Ginger exosome-like nanoparticles (GELNs) attenuated the pathogenicity of P. gingivalis. This is because they reduce the expression of genes of this bacteria relationated with adhesion, hemagglutinin, proteinases, transcription factors and important components of the membrane. Consequently, GELNs can have anti-biofilm as well as antibacterial activity against P. gingivalis. Further, GELNs because of their natural source can be inexpensive produced. Therefore, this research will provide valuable information on the efficacy and applicability of GELNs to treat and prevent AD. The results of this research will guide the necessary conceptual or methodological adjustments, as well as the next steps for an exhaustive characterization and preclinical evaluation of this new promising therapeutic. |
