Insights into the microbial community of an enhanced biological phosphorus removal plant

Abstract: The biological phosphorus removal (EBPR) process relies mainly on polyphosphate accumulating organisms (PAOs). Nowadays their characterization still remains a challenge due to the complexity of wastewater communities and the lack of a single methodology that accurately identifies and quantifies these organisms. Fluorescence in situ hybridization enables identification/quantification of specific targeted groups, whereas next sequencing generation techniques provide a broader taxonomic overview of the microbial community. However, across literature there are significant discrepancies between these methodologies results, highlighting the need of a systematic approach reliable for PAO characterization. In this study three bacteria genera exhibiting PAO phenotype are evaluated. The three show differences in their relative abundances comparison, being Ca. Accumulibacter the most divergent (abundances <1% in FISH vs. >28% in NGS).
